Abstract

Introduction. In recent years, the attention of researchers has been drawn to the role of so-called ectopic taste receptors, located outside the canonical localization, in particular bitter taste receptors (Tas2R), in the regulation of cellular functions in bronchial asthma (BA). The aim of the study was to analyze the association of plasma levels of Tas2R1 with clinical and functional characteristics of different variants of BA. Materials and methods. Ten practically healthy individuals, 36 patients with allergic BA (ABA) and 20 patients with non-allergic BA (NABA) were examined. The level of Tas2R1 expression in the blood serum was determined by the enzyme immunoassay. Results. The blood plasma levels of bitter taste receptors Tas2R1 in different variants of BA differ significantly. In NABA, this level is significantly higher than in ABA. A direct correlation between Tas2R1 and the neutrophil content was found in ABA, and an inverse correlation with the number of monocytes in the peripheral blood in NABA. The relationship between Tas2R1 and the number of monocytes in sputum is positive in ABA. In NABA, both the content of macrophages and bronchial epithelium directly correlates with the level of Tas2R1. In ABA, a direct relationship between the Tas2R1 receptor and STAT6, STAT4, SOCS1, SOCS3 was found. A direct correlation between the level of the Tas2R1 receptor and airway resistance was revealed. Discussion. In general, it was found that the levels of the Tas2R1 receptor in blood plasma in different variants of BA differ significantly, namely, in NABA this level is significantly higher than in ABA. In ABA, a predominant correlation dependence of the Tas2R1 receptor on the negative regulator SOCS1 was revealed. Conclusions. Correlation analysis of Tas2R1 levels in different variants of BA revealed features related to the correlation with the cellular composition of peripheral blood and sputum, as well as with transcription factors regulating allergic inflammation.
